"Bendamustine is a novel agent consisting of a mechlorethamine (nitrogen mustard) group, a benzimidazole ring, and a butyric acid side chain. In vitro studies demonstrate rapid production of DNA crosslinks and strand breaks after bendamustine exposure. In addition to direct DNA damage and apoptosis, other mechanisms include **inhibition of mitotic checkpoints** and induction of mitotic catastrophe. These characteristics may explain the activity of bendamustine in drug-resistant cancer cells and refractory lymphoma patients. Benzimidazole acts as a purine antagonist in experimental models; the contribution of this structure to the overall antitumor activity of bendamustine is unknown."
